Unlike phage display, yeast display operates in a eukaryotic environment, which supports proper protein folding, post-translational modification, and surface translocation. Consequently, yeast-displayed antibodies resemble their native human counterparts more closely. By using flow cytometry (FACS), scientists can precisely screen large libraries of antibodies displayed on yeast, significantly improving the discovery process.

This method yields up to 2×10⁸ transformants per microgram of ligation mixture, which facilitates the generation of high-diversity libraries. Our Approach

At Rx Biosciences, we actively monitor antibody maturation through yeast display screening. The yeast-expressed antibodies undergo binding with their target antigen under gradually increasing stringency. Using Fluorescence-Assisted Cell Sorting (FACS), we then isolate yeast cells that bind strongly to the antigen. Afterward, selected antibodies are purified via affinity chromatography for further study.
