Custom PhIP-Seq Service for Profiling of Autoantibodies

Rx Biosciences offers a Custom PhIP-Seq service (Phage ImmunoPrecipitation Sequencing) that profiles autoantibodies in patient serum using phage display technology. In this process, we prepare a phage display library composed of predesigned oligopeptides, each 50 to 90 amino acids long, using a modified T7 Select vector available at Rx Biosciences. Next, we screen the library with a pool of antibodies directly isolated from a patient diagnosed with an autoimmune disorder.

After hybridization, Protein A/G resin selectively captures the antibody-phage complexes, and we amplify them using PCR. In the first round of PCR, we amplify and purify the library inserts. Then, during the second PCR round, we attach indexes to the various samples generated during the screening process. These indexed samples are subsequently sequenced using MiSeq technology.

Once sequencing is complete, we demultiplex, align, and analyze the FastQ files to determine the enrichment of patient-specific autoantibodies. Overall, PhIP-Seq experiments are highly effective for investigating environmental exposures, allergies, evolving autoimmune conditions, and responses to immunomodulatory therapies.
