Full Length cDNA

At Rx Biosciences, we specialize in the construction of high-quality full-length cDNA libraries using selectively captured full-length mRNAs. Our proprietary technology leverages a novel reverse transcriptase enzyme capable of accurately transcribing RNA molecules longer than 15 kb, overcoming challenges commonly faced with long and GC-rich RNA templates. The synthesis process begins with an oligo dT primer to initiate the first strand cDNA synthesis, ensuring the capture of the polyadenylated tail of mRNAs. Both the first and second strands are synthesized using advanced chemistry designed to abolish secondary structures and minimize interruptions that are typically caused by high GC-content regions in the RNA template. This approach guarantees the production of comprehensive and faithful cDNA copies of the entire transcriptome, which is essential for downstream applications such as gene expression analysis, cloning, and functional studies.

Our Approach

We generate full-length cDNA libraries starting from micro-quantities of total RNA extracted from hard-to-obtain samples including patient blood, laser capture microdissected (LCM) tissues, or fluorescence-activated cell sorting (FACS) sorted cells. These samples often present challenges due to their limited quantity and quality, but our optimized protocols allow for effective library construction without compromising transcript completeness. The final libraries are delivered cloned in phage-resistant, high-efficiency E. coli competent cells, ensuring stable propagation and high yields. This method enables researchers to work with rare or precious samples while still obtaining robust, full-length cDNA libraries that accurately represent the original RNA population. Whether for transcriptome profiling, isoform discovery, or gene cloning, our libraries provide a reliable foundation for diverse molecular biology applications.
